Overview

GC4711 (rucosopasem manganese) is a highly selective, small molecule superoxide dismutase (SOD) mimetic being developed by Galera Therapeutics. It is designed to be used in combination with stereotactic body radiation therapy (SBRT) for the treatment of non-small cell lung cancer (NSCLC) and other solid tumors. The drug works by rapidly converting superoxide radicals, which are generated in high bursts during radiation therapy, into hydrogen peroxide. Because cancer cells often have higher baseline levels of reactive oxygen species and lower antioxidant capacity compared to normal cells, the additional hydrogen peroxide generated by GC4711 selectively promotes cancer cell death. Simultaneously, GC4711 is intended to protect normal lung tissue from radiation-induced injury. GC4711 is a second-generation SOD mimetic, optimized for shorter infusion times compared to its predecessor, avasopasem manganese. It is currently being evaluated in the GRECO-1 Phase 1/2 clinical trial for patients with centrally located or large, node-negative NSCLC.
