GC511B is an autologous, lentiviral gene-modified chimeric antigen receptor T-cell (CAR-T) therapy designed to target Delta-like ligand 3 (DLL3). Developed by AstraZeneca following its acquisition of Gracell Biotechnologies, GC511B is being investigated for the treatment of relapsed or refractory small cell lung cancer (SCLC). DLL3 is a Notch ligand that is highly overexpressed on the surface of SCLC cells and other neuroendocrine tumors but has minimal expression in healthy tissues, making it an ideal target for cellular immunotherapy. The therapy utilizes the patient's own T cells, which are genetically engineered to express a chimeric antigen receptor specific for DLL3, enabling them to selectively recognize and eliminate DLL3-positive tumor cells. GC511B is currently being evaluated in Phase 1 clinical trials to determine its safety, feasibility, and preliminary antitumor activity.
