Overview

GCC19CAR-T is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of metastatic colorectal cancer (mCRC). It is the first clinical candidate from the CoupledCAR solid tumor platform and consists of a mixture of autologous CAR T cells transduced with lentiviral vectors encoding CARs targeting both guanylate cyclase-C (GCC), a mucosal marker highly expressed in colorectal cancers, and CD19. The dual-targeting approach aims to enhance proliferation, activation, and persistence of CAR T cells against solid tumors by leveraging CD19-targeted signaling to boost anti-GCC activity. In phase 1 trials in China and the United States, GCC19CAR-T has demonstrated manageable safety—primarily cytokine release syndrome and diarrhea—and promising antitumor activity in heavily pretreated mCRC patients[1][2][3][4][5][6]. The therapy is being developed by Innovative Cellular Therapeutics.
