Overview

GCC4001 + rituximab is a combination therapy consisting of allogeneic, cord blood-derived naive natural killer (NK) cells (GCC4001, also known as AB-101) and the anti-CD20 monoclonal antibody rituximab. Developed by GC Cell (Yongin Cell Therapy Center), the therapy is designed to treat CD20-positive B-cell malignancies and certain autoimmune diseases. The mechanism of action leverages antibody-dependent cellular cytotoxicity (ADCC), where the NK cells recognize the Fc portion of rituximab bound to CD20-expressing target cells via their CD16a receptors, leading to enhanced cytotoxic killing of malignant or pathogenic B cells. It is currently being evaluated in Phase 1 clinical trials for relapsed or refractory B-cell non-Hodgkin lymphoma and is under investigation for refractory rheumatoid arthritis and other autoimmune conditions.
