Overview

GCM-101 is an engineered herpes simplex virus type 1 (HSV-1)-based oncolytic virus developed by Gencellmed for the treatment of various solid tumors, including ovarian, prostate, pancreatic, breast, lung, and colon cancers. The virus utilizes a "Double Retargeting Technology" involving an HVEM-scFvEpCAM adapter and a modified gH-scFvEpCAM to specifically target Epithelial Cell Adhesion Molecule (EpCAM)-expressing tumor cells. To ensure tumor-selective replication, the virus is controlled by the human telomerase reverse transcriptase (hTERT) promoter. Additionally, GCM-101 is engineered to secrete immunostimulatory cytokines, specifically Interleukin-12 (IL-12), Interleukin-7 (IL-7), and Fms-related tyrosine kinase 3 ligand (FLT3LG), to enhance the anti-tumor immune response. It is currently in preclinical development.
