Overview

GcMAF (Gc protein-derived macrophage activating factor) is a biologically active glycoprotein produced by the enzymatic modification of vitamin D-binding protein (also known as Gc or group-specific component protein). It acts as a potent activator of macrophages—immune cells responsible for detecting and destroying pathogens and abnormal cells. The mechanism involves deglycosylation of the precursor vitamin D-binding protein to generate an active form that stimulates phagocytic activity, superoxide generation, anti-angiogenic effects, dendritic cell maturation, and antitumor immune responses. While it has been promoted for cancer immunotherapy and other chronic diseases such as HIV and hepatitis C by some clinics (notably in Japan), there is no robust clinical evidence supporting its efficacy for these indications. Several generations of GcMAF have been developed using different manufacturing processes; newer formulations claim higher concentration and stability[2][3][7]. Clinical trials are ongoing to assess its safety and efficacy in chronic inflammatory diseases and metastatic breast cancer[6][8].
