Drug intelligence / Profile preview
GCP-rCpa5 is a preclinical recombinant protein subunit vaccine candidate being developed for the prevention of Valley Fever (coccidioidomycosis), a potentially fatal respiratory fungal disease caused by *Coccidioides* species (*Coccidioides immitis* and *Coccidioides posadasii*). The vaccine consists of the engineered recombinant *Coccidioides* polypeptide antigen 5 (rCpa5), which contains multiple T-cell epitopes for prevalent human MHC-II alleles (such as HLA-DR4 and HLA-DR3) to enhance solubility and broaden HLA coverage compared to its predecessor rCpa1. The rCpa5 antigen is formulated with glucan-chitin particles (GCPs) as an adjuvant and delivery system. GCP-rCpa5 stimulates protective Th1 and Th17 immune responses and has demonstrated efficacy in reducing fungal burden and improving survival in animal models of *Coccidioides* infection.
