GCT-007 + PD-L1 inhibitor is a preclinical-stage combination therapy developed by Global Cancer Technology. The regimen combines GCT-007 (also known as GCT.GLIO.1), a selective, brain-penetrant small molecule inhibitor of Phosphoinositide 3-kinase (PI3K), with an undisclosed PD-L1 checkpoint inhibitor. This combination is designed to enhance anticancer activity by modulating immune cell function and converting immunologically cold tumors into hot tumors, thereby improving immune recognition and infiltration. Preclinical studies in glioblastoma models have demonstrated significant tumor elimination and superiority over other PI3K inhibitors like paxalisib. The therapy is currently being investigated for the treatment of glioblastoma (GBM), diffuse midline glioma (DIPG), and tamoxifen-resistant breast cancer.
