GCT-008 is a potent, selective, and orally bioavailable small molecule inhibitor of VPS34 (vacuolar protein sorting 34, also known as phosphatidylinositol 3-kinase catalytic subunit type 3), a lipid kinase that plays a key role in autophagy initiation and lysosomal trafficking. Developed by Global Cancer Technology, Inc., it is being explored preclinically for oncology indications, particularly neoplasms such as glioblastoma (brain tumors) and triple-negative breast cancer, where VPS34 inhibition may overcome resistance to PI3K/AKT/mTOR pathway inhibitors by disrupting cytoprotective autophagy.
