GD-07 is a preclinical small molecule developed by Gudui BioPharma for the treatment of multiple myeloma. It acts as a molecular glue targeting the cereblon (CRBN)/IKZF1/IKZF3 axis. By binding to the CRBN E3 ubiquitin ligase complex, GD-07 facilitates the recruitment, ubiquitination, and subsequent proteasomal degradation of the transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). These proteins are essential for the survival of malignant plasma cells, and their degradation leads to cell cycle arrest and apoptosis. This mechanism of action is analogous to that of immunomodulatory imide drugs (IMiDs), but GD-07 is being developed as a next-generation agent with potentially improved efficacy or safety profiles.
