GD-09 is a preclinical small molecule drug candidate developed by Gudui BioPharma for the treatment of multiple myeloma. It functions by modulating the CRBN/IKZF1/IKZF3 axis, a well-established pathway in B-cell malignancies. As a molecular glue or cereblon E3 ubiquitin ligase modulator (CELMoD), GD-09 facilitates the interaction between the Cereblon (CRBN) E3 ligase complex and the transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). This recruitment leads to the polyubiquitination and subsequent proteasomal degradation of IKZF1 and IKZF3, which are critical for the survival and proliferation of myeloma cells. By depleting these essential proteins, GD-09 induces apoptosis and inhibits tumor growth in multiple myeloma models.
