Gd-DOTA-ob5 is a novel MRI contrast agent specifically designed to target and visualize oligomeric amyloid-beta (oAβ) deposits in the brain for early diagnosis and research in Alzheimer’s disease. It consists of a gadolinium (Gd)-dodecane tetraacetic acid (DOTA) chelate covalently attached to a DNA aptamer called ob5, which was developed using SELEX to bind with high specificity and affinity to oAβ, a key pathogenic species implicated in early-stage Alzheimer’s disease. Unlike conventional gadolinium-based contrast agents, Gd-DOTA-ob5 selectively accumulates at sites with oAβ, enhancing T1-weighted MRI signals in animal models of Alzheimer’s disease. Its development aims to facilitate early, non-invasive detection and monitoring of oAβ pathology, and enable evaluation of therapeutic interventions in preclinical models[1][2][4].
