gD-HPV-16 is a therapeutic vaccine candidate developed by GSK for the treatment of HPV-16-related conditions, including cervical intraepithelial neoplasia (CIN) and cervical cancer. The vaccine is a recombinant fusion protein consisting of a detoxified version of the Human Papillomavirus type 16 (HPV-16) E7 oncoprotein fused to the glycoprotein D (gD) of Herpes Simplex Virus type 1 (HSV-1). The E7 protein is a critical target for therapeutic intervention as it is constitutively expressed in HPV-transformed cells and is essential for the maintenance of the malignant phenotype. The gD component serves as a molecular adjuvant and signal sequence that directs the fusion protein to the endosomal/lysosomal compartments of antigen-presenting cells, thereby enhancing MHC class II presentation and stimulating a potent T-cell mediated immune response. Formulated with GSK's AS02 adjuvant system, the vaccine was designed to induce cellular immunity capable of identifying and eliminating HPV-infected cells. Clinical development of gD-HPV-16 has been discontinued following early-phase trials.
