Overview

GD2-28z CAR T cells are an adoptive cell therapy consisting of T cells engineered to express a chimeric antigen receptor (CAR) targeting the disialoganglioside GD2. The CAR construct typically utilizes the 14G2a single-chain variable fragment (scFv) for antigen recognition, coupled with a CD28 costimulatory domain and a CD3-zeta (ζ) signaling domain. GD2 is highly expressed on various neuroectoderm-derived tumors, including neuroblastoma, osteosarcoma, melanoma, and certain brain tumors like diffuse intrinsic pontine glioma (DIPG). The inclusion of the CD28 domain is intended to enhance T cell expansion and persistence upon antigen encounter. This specific construct has been widely used in academic research and early-phase clinical trials, notably by investigators at the National Cancer Institute and Stanford University.
