Overview

GD2-CAR-expressing autologous T-lymphocytes is an investigational chimeric antigen receptor (CAR) T-cell therapy developed by the National Cancer Institute (NCI) for the treatment of GD2-positive pediatric solid tumors, including osteosarcoma and neuroblastoma. The therapy involves the genetic modification of a patient's own T cells to express a CAR specific for the disialoganglioside GD2, which is highly expressed on the surface of these tumors but has limited expression in normal tissues. Once re-infused into the patient, typically following lymphodepleting chemotherapy with cyclophosphamide and fludarabine, these engineered cells recognize and kill GD2-expressing cancer cells. Clinical evaluation in Phase I trials has focused on safety, persistence, and the ability of the cells to traffic to tumor sites, showing a manageable safety profile with limited cytokine release syndrome.
