Overview

GD2-CAR-modified tri-virus-specific cytotoxic T-lymphocytes are an investigational adoptive cell therapy designed to treat GD2-positive solid tumors, most notably neuroblastoma and osteosarcoma. This specialized CAR-T therapy utilizes a "virus-specific T-cell" (VST) backbone, where the T-lymphocytes are selected for their endogenous T-cell receptor (TCR) specificity against three common viruses: Epstein-Barr virus (EBV), Cytomegalovirus (CMV), and Adenovirus (Adv). These VSTs are then genetically engineered with a chimeric antigen receptor (CAR) targeting the disialoganglioside GD2. The primary mechanism involves the CAR targeting tumor cells expressing GD2 for cytotoxic killing. The secondary, unique mechanism leverages the tri-virus-specific TCRs to provide the T-cells with periodic stimulation from latent or active viral antigens in the patient, which is intended to improve the survival, expansion, and long-term persistence of the T-cells in the immunosuppressive microenvironment of solid tumors.
