Overview

GD2 CAR T refers to a class of chimeric antigen receptor (CAR) T-cell therapies engineered to recognize and eliminate cancer cells expressing the disialoganglioside GD2. GD2 is a tumor-associated antigen highly expressed on neuroblastoma, diffuse midline gliomas (including DIPG), glioblastoma, and certain other solid tumors, while maintaining restricted expression in normal tissues. These therapies typically involve the modification of autologous T cells with a CAR construct containing a GD2-specific single-chain variable fragment (scFv), often derived from the 14G2a antibody, linked to co-stimulatory (e.g., 4-1BB or CD28) and signaling (CD3ζ) domains. Clinical development is being led by academic institutions such as Stanford University and Bambino Gesù Children's Hospital, showing promise in pediatric and adult neuro-oncology and refractory neuroblastoma.
