GD2-iCAR-PBT is an autologous cell therapy in which patient-derived peripheral blood T cells are genetically engineered to express a third-generation chimeric antigen receptor (CAR) targeting the tumor-associated antigen GD2. This construct includes CD3ζ, CD28, and OX40 signaling domains, features a 14g2a single-chain variable fragment (scFv) for GD2 binding, and incorporates an inducible caspase 9 (iCasp9) suicide gene for safety. The product is manufactured at the point of care and delivered via intravenous infusion. It is being studied primarily for metastatic melanoma and other GD2-positive solid tumors, with the intent to offer an immunotherapeutic alternative in patients for whom conventional therapies (such as checkpoint inhibitors) are insufficient. The therapy was initially developed by Sydney Children's Hospitals Network and is in Phase 1 clinical trials.
