GD2-IL18-CAR-T is an armored chimeric antigen receptor (CAR) T-cell therapy developed by researchers at the University of Münster. It is designed to target the disialoganglioside GD2, a surface antigen highly expressed in various pediatric and adult solid tumors, including neuroblastoma, osteosarcoma, and Ewing sarcoma. To enhance the therapeutic efficacy and metabolic fitness of the T cells within the immunosuppressive tumor microenvironment, these CAR-T cells are engineered to constitutively secrete Interleukin-18 (IL-18). IL-18 is a proinflammatory cytokine that promotes Th1-type immune responses, activates natural killer (NK) cells, and improves the persistence and anti-tumor activity of the CAR-T cells. This dual-action approach aims to overcome the limitations of conventional CAR-T therapies in solid tumors by combining direct antigen-targeted cytotoxicity with localized cytokine-mediated immune modulation.
