GD2-SADA-177Lu-Proteus is a next-generation radiopharmaceutical developed by Y-mAbs Therapeutics using the Self-Assembling Disassembling Antibody (SADA) Pre-targeted Radioimmunotherapy (PRIT) platform. The therapy utilizes a two-step approach: first, a bispecific SADA protein targeting the ganglioside GD2 and a DOTA-chelated radioligand is administered. This protein self-assembles into tetramers to bind GD2-expressing tumor cells and then clears rapidly from the blood as monomers. In the second step, a small molecule radioligand carrying the beta-emitting isotope Lutetium-177 (177Lu) is administered, which binds to the pre-targeted SADA protein at the tumor site. This mechanism is designed to deliver high doses of localized radiation to tumors while minimizing systemic exposure and off-target toxicity. It is currently in Phase 1 clinical development for GD2-positive solid tumors, including small cell lung cancer, sarcoma, melanoma, and neuroblastoma.
