GD2-specific 4SCAR-T cells are a fourth-generation safety-designed chimeric antigen receptor T cell therapy that targets the GD2 antigen. These engineered T cells incorporate multiple intracellular signaling domains including CD28/4-1BB/CD3ζ-iCasp9 for enhanced activation and safety. The therapy includes an inducible caspase-9 (iCasp9) safety switch that allows for controlled elimination of the CAR-T cells if needed. The cells are genetically modified using lentiviral vectors and are designed to treat GD2-positive tumors including glioblastoma and neuroblastoma. The fourth-generation design aims to improve persistence and reduce toxicity compared to earlier CAR-T cell generations.
