Overview

GD2.CAR-TVS-CTL is a chimeric antigen receptor T cell therapy targeting the GD2 antigen, which is highly expressed in certain solid tumors including diffuse midline gliomas (DMG), diffuse intrinsic pontine glioma (DIPG), glioblastoma, and neuroblastoma[1][3][4][5][6][7]. The drug consists of autologous or allogeneic T cells that are genetically engineered to express a GD2-targeted CAR, enabling them to recognize and kill GD2-expressing tumor cells. Administration requires prior lymphodepleting chemotherapy, and the therapy is investigated for intravenous as well as intracerebroventricular delivery[1][3][6]. GD2.CAR-TVS-CTL is designed as a living drug with the capacity for expansion and persistence in the patient, offering the potential for sustained anti-tumor activity. In clinical trials, GD2-CAR T cell therapy demonstrates significant tumor regression, neurological improvements, and durable remissions in pediatric and young adult patients with H3K27M-mutant DMG/DIPG and high-risk neuroblastoma[1][2][3][4]. The mechanism of action is targeted cytotoxicity via T cell engagement of GD2+ tumor cells, leading to their destruction. Toxicities observed include tumor inflammation-associated neurotoxicity and cytokine release syndrome, generally manageable under intensive monitoring[1][3][6].
