GD55 is a novel oncolytic adenovirus engineered to selectively replicate in and destroy cancer cells, particularly those expressing high levels of the Golgi protein GOLPH2 (also known as GP73). GD55 is regulated by the GOLPH2 promoter, enabling preferential targeting and oncolytic activity against hepatocellular carcinoma (HCC) and other tumor types expressing GOLPH2. Preclinical studies have demonstrated potent cytotoxic effects on liver cancer stem-like cells and prostate cancer stem cell-like cells both in vitro and in vivo, with limited effects on normal cells. The virus achieves these effects by replicating within cancer cells, inducing cell lysis, and promoting apoptosis, which results in tumor regression and reduced cancer stem cell populations. GD55 was developed to improve upon earlier oncolytic adenoviruses, such as ZD55, by enhancing targeting and replication efficiency in tumor cells.[1][2][4]
