GDC-0134 is a potent, selective, orally available, brain‐penetrant small-molecule inhibitor of dual leucine zipper kinase (DLK). DLK regulates the c-Jun N-terminal kinase (JNK) pathway involved in axon degeneration and neuronal apoptosis following injury. By inhibiting DLK, GDC-0134 aims to block JNK activation and protect neurons from degeneration. It was developed by Genentech for the treatment of amyotrophic lateral sclerosis (ALS), with the goal of slowing disease progression by preventing motor neuron loss[1][3][5].
