GDC-0334

GDC-0334 is a novel, orally administered small molecule that acts as a selective antagonist of the transient receptor potential cation channel subfamily A member 1 (TRPA1). It was developed by Genentech for the treatment of TRPA1-mediated diseases such as pain, asthma, and other inflammatory or respiratory conditions. Preclinical studies demonstrated that GDC-0334 inhibits TRPA1 function on airway smooth muscle and sensory neurons, reducing cough and allergic airway inflammation in animal models. The drug binds to a deep pocket within the transmembrane domain of TRPA1, stabilizing its closed state and preventing channel activation. Clinical development reached phase 1 trials in healthy volunteers for indications including severe asthma; however, further development was discontinued.
