Overview

**GDC-0425 + gemcitabine** is an investigational combination therapy composed of GDC-0425—a selective, orally bioavailable small-molecule checkpoint kinase 1 (Chk1) inhibitor—and gemcitabine, a cytotoxic nucleoside analog[1][4]. GDC-0425 is designed to potentiate the effects of DNA-damaging chemotherapy by inhibiting Chk1, thereby overriding cell-cycle checkpoints and genome repair, sensitizing tumor cells to gemcitabine[2][4]. Gemcitabine acts primarily as an antimetabolite, inhibiting DNA synthesis and promoting cell death. The combination has been studied primarily in advanced or refractory solid tumors and lymphoma, with the rationale that Chk1 inhibition may enhance the cytotoxicity of agents like gemcitabine[1][3][4].
