GDC-0575 is a highly selective oral small-molecule inhibitor of checkpoint kinase 1 (Chk1), a protein kinase that regulates the tumor cell response to DNA damage. By specifically binding to and inhibiting Chk1, GDC-0575 disrupts Chk1-dependent cell cycle arrest in the S and G2/M phases. This inhibition prevents cancer cells from repairing DNA damage before mitosis, leading to increased tumor cell death when combined with DNA-damaging chemotherapies such as gemcitabine or cytarabine. Preclinical studies have shown that GDC-0575 results in tumor shrinkage and growth delay in xenograft models. The drug was invented by Array BioPharma and licensed to Genentech for clinical development and commercialization[1][2][4][7].
