Overview
GDC-3280 is an orally bioavailable small molecule antagonist of Toll-like receptor 7 (TLR7) and Toll-like receptor 8 (TLR8), developed by Genentech. These endosomal receptors are key components of the innate immune system, recognizing single-stranded RNA and triggering the production of pro-inflammatory cytokines and type I interferons. Dysregulated TLR7/8 signaling is implicated in the pathogenesis of autoimmune and fibrotic diseases, including idiopathic pulmonary fibrosis (IPF) and systemic sclerosis. By blocking these receptors, GDC-3280 aims to reduce the inflammatory and fibrotic drivers of disease progression. The compound has been evaluated in Phase 1 clinical trials to assess its safety, tolerability, and pharmacokinetics in healthy volunteers and patients with systemic sclerosis.
