Overview

GDC-8264 is a potent, selective, and reversible small molecule inhibitor of receptor-interacting protein kinase 1 (RIP1). RIP1 is a key regulator of inflammatory responses and cell death pathways such as apoptosis and necroptosis. By inhibiting RIP1 kinase activity, GDC-8264 aims to reduce inflammation and tissue damage in diseases where these processes are implicated. The drug is being developed by Genentech for the treatment of acute graft-versus-host disease (aGVHD) following allogeneic hematopoietic stem cell transplantation and for the prevention of cardiac surgery-associated acute kidney injury (AKI). It has also been studied in healthy volunteers to assess safety, pharmacokinetics, and pharmacodynamics[2][5][6][8].
