Overview

**GDI-4405** is an oral small molecule antiviral drug candidate developed by the Global Health Drug Discovery Institute (GHDDI) targeting the 3CL (3C-like or main) protease of SARS-CoV-2. It demonstrates superior potency to Pfizer's nirmatrelvir (the active ingredient in Paxlovid) in inhibiting viral replication across multiple assays, including those using primary human airway epithelial cells infected with live SARS-CoV-2 and the Delta variant, with good oral bioavailability in rats and cynomolgus monkeys, an excellent safety profile in animal toxicology, and significantly lower manufacturing costs. Its efficacy is unaffected by SARS-CoV-2 mutations like Omicron due to the conserved nature of the 3CL protease target, positioning it as a potential treatment and preventive for evolving COVID-19 variants.[1][2]
