Overview

gdL-28Z is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the constant region of the gamma-delta (γδ) T-cell receptor (TCR). Developed by researchers at the National Cancer Institute (NCI), the construct utilizes a specific single-chain variable fragment (scFv) designated 'gdL' for antigen recognition, paired with a CD28 hinge and transmembrane domain, a CD28 costimulatory domain, and a CD3ζ activation domain. It is intended for the treatment of aggressive γδ T-cell malignancies, such as hepatosplenic T-cell lymphoma and T-cell acute lymphoblastic leukemia (T-ALL), as well as potential applications in autoimmune diseases. In preclinical studies, gdL-28Z demonstrated antigen-specific activation and cytotoxicity against γδ TCR-expressing cell lines, although it showed lower potency compared to the related MGDL-28Z construct.
