GDPD5-shRNA is a short hairpin RNA (shRNA) gene therapy construct designed to silence the expression of glycerophosphodiester phosphodiesterase domain containing 5 (GDPD5). GDPD5 is an enzyme with glycerophosphocholine phosphodiesterase (GPC-PDE) activity that is upregulated in breast cancer cells, leading to altered choline phospholipid metabolism characterized by low levels of glycerophosphocholine (GPC) and high levels of phosphocholine (PC). By knocking down GDPD5 expression, GDPD5-shRNA restores GPC levels and reduces PC levels, shifting the metabolic profile of breast cancer cells toward a less malignant phenotype. Preclinical studies in breast cancer models, including MCF-7 and MDA-MB-231 cell lines and orthotopic tumor xenografts, have demonstrated that GDPD5 silencing reduces tumor cell proliferation, migration, invasion, and in vivo tumor growth.
