GE11-liposomal CD73 siRNA is an experimental EGFR-targeted nanoparticle formulation designed to silence the expression of CD73 (ecto-5'-nucleotidase) in the tumor microenvironment. The formulation utilizes the GE11 peptide, which binds specifically to the Epidermal Growth Factor Receptor (EGFR), to facilitate targeted delivery of siRNA-loaded nanoliposomes to tumor cells. By knocking down CD73, the drug reduces the conversion of extracellular AMP to immunosuppressive adenosine, thereby enhancing anti-tumor immune responses, increasing CD8+ T-cell activity, and reducing regulatory T-cell (Treg) infiltration. It has been studied primarily in preclinical models of metastatic breast cancer, often in combination with chemotherapy agents like liposomal doxorubicin.
