**GE11-Ori-Se NPs** are a preclinical EGFR-targeted nanomedicine comprising the diterpenoid anticancer compound **oridonin** loaded into selenium nanoparticles functionalized with the EGFR-binding GE11 peptide. The nanoparticles preferentially enter EGFR-overexpressing cancer cells, accumulate in lysosomes, and release oridonin more readily under acidic conditions. In esophageal cancer cell and mouse xenograft studies, the formulation induced reactive oxygen species generation and mitochondria-associated apoptosis, suppressed EGFR-associated PI3K/AKT and Ras/Raf/MEK/ERK signaling, reduced tumor angiogenesis, and inhibited tumor growth. It was developed as an intravenously administered candidate for EGFR-overexpressing esophageal cancer.
