Overview

GE2 is a designation used for two distinct experimental therapeutic candidates. The first is a **recombinant human fusion protein** (also known as Gamma-Epsilon 2) developed by **Tunitas Therapeutics** for the treatment of asthma and allergic diseases. This molecule is designed to co-aggregate the high-affinity IgE receptor (**FcεRI**) and the inhibitory IgG receptor (**FcγRIIb**) on the surface of mast cells and basophils. By bringing these receptors together, GE2 triggers an inhibitory signaling pathway (via SHIP recruitment to the ITIM of FcγRIIb) that suppresses the release of inflammatory mediators like histamine. The second candidate referred to as GE2 is a **peptide-drug conjugate** consisting of **glucagon-like peptide-1 (GLP-1)** linked to **estradiol**. This dual-action agent acts as an agonist for both the GLP-1 receptor and the estrogen receptor, aiming to provide metabolic benefits (such as weight loss and glucose regulation) and neuroprotective effects (such as enhanced neurogenesis and cognitive improvement) while minimizing the systemic side effects of estrogen by targeting it to GLP-1 receptor-expressing tissues.
