GE23 is a novel hybrid homo-aza (lactam) steroidal antimetabolite designed for the treatment of gastrointestinal malignancies, specifically pancreatic and colorectal cancers. It belongs to a class of steroidal conjugates developed to enhance the selectivity and toxicokinetics of conventional antimetabolites. GE23 acts as a dual inhibitor of thymidylate synthase (TS) and dihydrofolate reductase (DHFR), enzymes critical for DNA synthesis and repair. By inhibiting these targets, GE23 induces cell cycle arrest and apoptosis in cancer cells. Preclinical studies have demonstrated its antiproliferative activity against various carcinoma cell lines, providing a rationale for its further evaluation as a next-generation therapeutic for resistant cancers.
