Overview

Gefapixant is a small molecule, orally administered selective antagonist of the P2X3 receptor (also known as P2RX3), developed for the treatment of refractory or unexplained chronic cough in adults. By blocking P2X3 receptors—ligand-gated ion channels found on sensory nerve fibers in the airway—gefapixant reduces cough reflex sensitivity triggered by extracellular ATP released during airway irritation or inflammation. The drug has demonstrated statistically significant reductions in cough frequency and improvements in quality of life measures compared to placebo at a dose of 45 mg twice daily. Taste-related adverse events are common and dose-dependent. Gefapixant was developed by Merck and is marketed under the brand name Lyfnua in some regions[1][2][3][4][6][8].
