Overview
Gefitinib is an oral small molecule inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. It blocks EGFR signaling pathways involved in cell proliferation and survival. 5-Fluorouracil (5-FU) is a pyrimidine analog that inhibits thymidylate synthase, disrupting DNA synthesis and function. Leucovorin enhances the binding of 5-FU to its target enzyme, increasing its cytotoxicity. Radiotherapy uses ionizing radiation to damage DNA in cancer cells and inhibit tumor growth. This combination has been investigated for synergistic effects in treating advanced or metastatic colorectal cancer and rectal cancer by targeting multiple mechanisms of tumor cell survival[1][4][6].
