Overview

Gefitinib + hydroxycamptothecin is an experimental combination therapy consisting of two small molecule anticancer agents. Gefitinib is a selective inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase. It blocks EGFR signaling by binding to its ATP-binding site and thereby inhibits downstream pathways involved in cell proliferation and survival. Hydroxycamptothecin (HCPT), also known as 10-hydroxycamptothecin, is a topoisomerase I inhibitor that induces DNA damage by stabilizing the cleavable complex between topoisomerase I and DNA during replication. The combination aims to exploit potential synergistic effects between EGFR inhibition and DNA damage induction for enhanced antitumor activity. Preclinical studies with similar combinations (e.g., gefitinib plus SN-38 or irinotecan) have shown schedule-dependent synergy in various cancer cell lines. This approach may be relevant for overcoming resistance mechanisms or enhancing efficacy in cancers such as non-small cell lung cancer (NSCLC) or colorectal cancer.
