This drug is a combination therapy of two targeted small molecule tyrosine kinase inhibitors used primarily in the treatment of advanced non-small cell lung cancer (NSCLC) with specific genetic alterations. Gefitinib is an epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor that targets EGFR mutations commonly found in NSCLC. Savolitinib is a highly selective oral MET tyrosine kinase inhibitor targeting the mesenchymal epithelial transition factor (c-MET), which is often amplified as a mechanism of resistance to EGFR inhibitors. The combination aims to overcome acquired resistance due to MET amplification in EGFR-mutated NSCLC patients who have progressed on prior EGFR-TKI therapy. Clinical studies, including phase Ib trials, have demonstrated acceptable safety profiles and promising antitumor activity, particularly in patients with MET amplification and EGFR mutation-positive NSCLC.
