Overview

GEM3-PSCA is a bispecific antibody designed to engage T-cells for the targeted treatment of cancers expressing prostate stem cell antigen (PSCA). It functions by simultaneously binding to CD3 on T-cells and PSCA on tumor cells, thereby redirecting T-cell cytotoxicity toward cancer cells. The drug was initially developed by GEMoaB Monoclonals and later advanced by AvenCell Europe. Its mechanism of action includes antibody-dependent cell cytotoxicity and immunologic cytotoxicity through T lymphocyte stimulation. Clinical development focused on patients with progressive disease after standard systemic therapy in PSCA-positive cancers such as prostate cancer, but also included other solid tumors like breast cancer, non-small cell lung cancer, pancreatic cancer, renal cancer, and urogenital cancers. Development has been discontinued following early-phase clinical trials[1][2][6].
