Overview

GemAGY is a novel gemcitabine analog developed to overcome the rapid metabolic deamination of gemcitabine in the blood. It was synthesized by chemical modification of gemcitabine hydrochloride at the N4 position with hydroxylamine. In preclinical studies, GemAGY demonstrated significantly higher cytotoxic activity against pancreatic cancer cell lines (MiaPaCa-2, BxPC3, and PANC-1) compared to gemcitabine, with lower IC50 values. The modification aims to improve metabolic stability and limit resistance mechanisms common in pancreatic cancer treatment. GemAGY works by inducing apoptosis, inhibiting cell migration, and causing cell cycle arrest in the G0/G1 phase.
