Drug intelligence / Profile preview
GemAGY-LnP is an EGFR-targeted immunoliposome encapsulating 4-N-hydroxylgemcitabine (GemAGY), a modified analog of gemcitabine designed for improved metabolic stability. The formulation utilizes the monoclonal antibody cetuximab as a targeting ligand to bind to the epidermal growth factor receptor (EGFR), which is frequently overexpressed in pancreatic cancer. By targeting EGFR, GemAGY-LnP enhances cellular uptake and accumulation of the cytotoxic payload within tumor tissues. Preclinical studies in MiaPaCa-2 spheroids and patient-derived xenograft (PDX) models have demonstrated significantly improved pharmacokinetic profiles, including increased half-life and area under the curve (AUC), as well as superior antitumor efficacy compared to standard gemcitabine hydrochloride.
