Overview

Gemcitabine + carboplatin + trastuzumab is a **three-drug combination regimen** used primarily in the treatment of metastatic breast cancer, particularly for patients whose tumors overexpress HER2/neu. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking and apoptosis, and trastuzumab is a humanized monoclonal antibody that targets the ErbB2 (HER2) receptor, blocking its ability to stimulate tumor cell growth and survival[3][5][7]. This combination has demonstrated synergistic antitumor activity, producing high response rates in HER2-positive metastatic breast cancer and activity in HER2-negative disease as well. Major toxicities are hematologic (notably myelosuppression); severe nonhematologic toxicity is uncommon[3][5][7]. The regimen is typically delivered intravenously in cycles and is regarded as a first-line therapy in clinical trials for metastatic breast cancer.
