Overview

Gemcitabine + cetuximab is a combination therapy used primarily in oncology, particularly for the treatment of certain solid tumors such as head and neck squamous cell carcinoma (HNSCC) and metastatic colorectal cancer. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, leading to cell cycle arrest in the S phase and apoptosis. Cetuximab is a recombinant chimeric human/mouse IgG1 monoclonal antibody that targets the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling pathways involved in tumor growth, survival, invasion, and angiogenesis. The combination has shown enhanced cytotoxic effects compared to either agent alone due to complementary mechanisms—gemcitabine induces S-phase accumulation while cetuximab causes G1 arrest—resulting in increased apoptosis of cancer cells[3][6]. Cetuximab was developed by ImClone Systems/Bristol Myers Squibb/Merck KGaA.
