Overview

Gemcitabine + cisplatin is a combination chemotherapy regimen that was evaluated by the NCIC Clinical Trials Group (now the Canadian Cancer Trials Group) in a Phase II clinical trial for patients with advanced, metastatic, or locoregionally recurrent malignant salivary gland tumors. Gemcitabine is a nucleoside analog that interferes with DNA synthesis by inhibiting ribonucleotide reductase and competing with cytidine for incorporation into DNA. Cisplatin (or its analog carboplatin, used in patients with contraindications to cisplatin) is a platinum-based antineoplastic agent that forms covalent DNA cross-links, leading to DNA damage and apoptosis. The trial (NCIC CTG HN.4) demonstrated an objective response rate of 24%, which did not meet the predefined efficacy criteria to warrant further investigation in this specific indication.
