gemcitabine + cisplatin + lenvatinib is a combination therapy regimen being evaluated for the treatment of advanced or unresectable intrahepatic cholangiocarcinoma. The regimen consists of lenvatinib, an oral multi-kinase inhibitor, combined with the chemotherapy doublet of gemcitabine and cisplatin (GC). Lenvatinib functions by inhibiting multiple receptor tyrosine kinases involved in angiogenesis and tumor growth, including VEGFR1-3, FGFR1-4, PDGFRα, RET, and KIT. Gemcitabine is a nucleoside metabolic inhibitor that disrupts DNA synthesis, while cisplatin is a platinum-based agent that forms DNA cross-links, leading to apoptosis. This specific combination is being investigated in clinical trials led by Shanghai Zhongshan Hospital to improve outcomes in patients with biliary tract cancers.
