Overview
Gemcitabine + cisplatin + paclitaxel is a combination chemotherapy regimen used primarily for the treatment of advanced or metastatic cancers, including non-small cell lung cancer (NSCLC) and biliary tract cancers. Each component has a distinct mechanism of action: - Gemcitabine is an antimetabolite that inhibits DNA synthesis by incorporating into DNA and inhibiting ribonucleotide reductase. - Cisplatin is a platinum-based compound that forms DNA crosslinks, leading to apoptosis. - Paclitaxel stabilizes microtubules and prevents their depolymerization, thereby inhibiting cell division. This combination has demonstrated feasibility and efficacy in clinical trials for advanced NSCLC with improved response rates and quality of life compared to historical controls. It is also being investigated in other solid tumors such as biliary tract cancer[2][5][3].
