This combination therapy regimen consists of the antimetabolite gemcitabine, the platinum-based alkylating agent cisplatin, and a monoclonal antibody targeting the Programmed Cell Death Protein 1 (PD-1) receptor. This triplet regimen has become a standard of care for the first-line treatment of recurrent or metastatic nasopharyngeal carcinoma (NPC). Gemcitabine and cisplatin provide cytotoxic debulking and potentially increase tumor immunogenicity, while the PD-1 antibody blocks the PD-1/PD-L1 pathway to restore T-cell mediated anti-tumor immune responses.
