This is a combination regimen consisting of two chemotherapeutic agents, gemcitabine and cisplatin, together with a programmed cell death protein 1 (PD-1) inhibitor. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, while cisplatin is a platinum-based compound that causes DNA crosslinking and apoptosis. The addition of a PD-1 inhibitor—such as nivolumab (Opdivo) or penpulimab—blocks the interaction between PD-1 on T cells and its ligands (PD-L1/PD-L2), thereby enhancing anti-tumor immune responses. This combination has been studied in various cancers including urothelial carcinoma and nasopharyngeal carcinoma, showing improved efficacy over chemotherapy alone in some settings[1][2].
